چکیده

The partition coefficient Kp was measured for a headgroup-labeled phospholipid (12:0,12:0)-N-(7-nitrobenz-2oxa-1,3-diazol-4-yl)-PE (12-NBD-PE), equilibrated between LUV of a series of phosphatidylcholines (PC). Fluorescence resonance energy transfer between the 12-NBD-PE and a headgroup-rhodamine-labeled PE was used to find the equilibrium concentration of the 12-NBD-PE in the different LUV. Reliable equilibrium concentrations were obtained by monitoring the approach to equilibrium starting from a concentration below and from a concentration above the ultimate values. Using (16:0,18:1 A9)-PC as the reference lipid, Kp ranged from a high value of 1.65 favoring (16:0,18:1 A9)-PC over (16:1 A9,16:1 A9)PC, to a low value of 0.90, favoring (22:11A 3,22:1A13)-PC over (16:0,18:1A9)-PC. The Kp values enabled calculation of the acyl chain contribution to the excess free energy of mixing for (1 2:0,12:0) acyl chains at infinite dilution in the Laf phase of PC having acyl chains of (16:0,18:1 A9), (16:1 A9,16:1 A9), (18:1 A9,18:1 A9), (18:1 A6,18:1 A6), (20:1 Al 1,20:1 Al 1), and (22:1 Al 3,22: 1 Al 3). (14:1 A9,14:1 A9)-PC was found to transfer so rapidly between LUV as to preclude reliable Kp measurement.

Shield as Signal: Lipopolysaccharides and the Evolution of Immunity to Gram-Negative Bacteria

According to the innate immunity concept [1], animals defend themselves from microbes by recognizing pathogen-associated molecular patterns. To detect many Gram-negative bacteria, animals use the CD14–MD-2– TLR4 receptor mechanism to recognize the lipid A moiety of the cell wall lipopolysaccharide (LPS). Lipid A is a glucosamine disaccharide that carries phosphates at positions 1 and 49 and usu...

Adsorption of mycoplasmavirus MV-L2 to Acholeplasma laidlawii: effects of changes in the acyl-chain composition of membrane lipids.

The enveloped mycoplasmavirus MV-L2 and its host Acholeplasma laidlawii JA1 were used to study the ways in which changes in the membrane lipid bilayer affect virus adsorption. The physical state of the membranes was altered by (i) using viruses and bacteria with different membrane lipid acyl-chain compositions, (ii) using incorporation of cholesterol, and (iii) changing the temperature. Adsorpt...

Role for phospholipid acyl chains and cholesterol in pulmonary infections and inflammation.

Bacterial and viral respiratory tract infections result in millions of deaths worldwide and are currently the leading cause of death from infection. Acute inflammation is an essential element of host defense against infection, but can be damaging to the host when left unchecked. Effective host defense requires multiple lipid mediators, which collectively have proinflammatory and/or proresolving...

Area per lipid and acyl length distributions in fluid phosphatidylcholines determined by (2)H NMR spectroscopy.

Deuterium ((2)H) NMR spectroscopy provides detailed information regarding the structural fluctuations of lipid bilayers, including both the equilibrium properties and dynamics. Experimental (2)H NMR measurements for the homologous series of 1, 2-diacyl-sn-glycero-3-phosphocholines with perdeuterated saturated chains (from C12:0 to C18:0) have been performed on randomly oriented, fully hydrated ...

Alterations in the metabolism of lipids in ischemia of the liver and kidney.

The metabolism of lipids in the ischemic liver has been examined in the attempt to define the cause of the previously described loss of phospholipid and to determine whether additional alterations occur that may be related to the disturbances in membrane function. With 3 hr of ischemia, 30% of the cellular phospholipid was lost when measured either as phosphate in a lipid extract of the whole l...

Molecular Insight into the Mutual Interactions of Two Transmembrane Domains of Human Glycine Receptor (TM23-GlyR), with the Lipid Bilayers

Appearing as a computational microscope, MD simulation can ‘zoom in’ to atomic resolution to assess detailed interactions of a membrane protein with its surrounding lipids, which play important roles in the stability and function of such proteins. This study has employed the molecular dynamics (MD) simulations, to determine the effect of added DMPC or DMTAP molecules on the structure of D...
